PENYUSUNAN BAHAN AJAR BIOTEKNOLOGI BERBASIS PENELITIAN PENGARUH MERKURI TERHADAP PERTUMBUHAN AZOTOBACTER DIISOLASI DARI RIZOZFER LEGUMINOCEAE

Abstract


Learning is the interaction of students with teachers and learning resources to gain positive knowledge, skills and values. This can be obtained by utilizing teaching materials as learning resources. The low value of daily biotechnology grade XII test, due to the low quality of teaching materials with conventional learning. The study aims to compose a Class-Based Biotechnology Research-Based Materials Effect of Mercury on the Growth of Azotobacter Bacteria Isolated from Rizozfer Leguminoceae. Observations performed bacterial sterilization, mercury resistance test, and Validation of teaching materials. The results of bacterial isolation from the Leguminoceae plant (peanuts, green beans, and long beans), followed by morphological observations to confirm the bacteria were Azotobacteria. Based on the isolation and characterization results, 3 bacteriaAzotobakter; AZKT, AZKH, and AZKP. The isolated bacteria were grown on mercury added media 0 ppm, 10 ppm, 15 ppm, and 20 ppm after bacteria were grown using bacterial counts using TPC. Based on the results of the study showed the amount of mercury concentration affect the growth of Azotobzcter bacteria. Mercury concentration 20 ppm growth of  K2M3 bacteria as much as 8,3x101 CFU/ml, 0 ppm growth of K1M0 bacteria as much as 109,7x101 CFU/ml highest bacteria count. The higher the mercury concentration, the less Azotobzcter bacteria grow. The results of the study were prepared as biotechnology materials of class XII SMA, and validated on 3 validator of Biology subject teachers and 2 lecturers of Biology Education. The results of validation of teaching materials showed the highest value of  V2 and V3 has the same value 37 or 84% and V1 has the lowest value of 32 or 72%, of all data validation results show more than 70% value which means quite valid (worth using)
